Biological Function Inhibitor of serine proteases. Its primary target is elastase, but it also has a moderate affinity for plasmin and thrombin. Irreversibly inhibits trypsin, chymotrypsin and plasminogen activator. The aberrant form inhibits insulin-induced NO synthesis in platelets, decreases coagulation time and has proteolytic activity against insulin and plasmin.; [Short peptide from AAT]: Reversible chymotrypsin inhibitor. It also inhibits elastase, but not trypsin. Its major physiological function is the protection of the lower respiratory tract against proteolytic destruction by human leukocyte elastase (HLE).

Background Alpha-1 Antitrypsin (alpha1AT) is a protease inhibitor of the serpin superfamily. It is generally known as serum trypsin inhibitor and is also referred to as alpha-1 proteinase inhibitor (alpha1PI) because it inhibits multiple proteases. It protects tissues from proteases of inflammatory cells, particularly neutrophil elastase, and has a reference range in blood of 1.5 – 3.5 gram/liter, but the concentration can rise significantly in cases of acute inflammation. In its absence (such as in alpha1AT deficiency), neutrophil elastase is free to break down elastin, which contributes to the elasticity of the lungs, resulting in respiratory complications such as emphysema, or COPD (chronic obstructive pulmonary disease) in adults and cirrhosis in adults or children.

Storage These antibodies are stable for at least one (1) year at -20°C to -70°C. Store product in appropriate aliquots to avoid multiple freeze-thaw cycles.
Dilution Instructions Dilute in PBS or medium that is identical to that used in the assay system.
Application Instructions These antibodies have been qualified for use in ELISA to detect human a1AT.
