Biological Function Catalyzes the reductive methylation of 2'-deoxyuridine 5'-monophosphate (dUMP) to thymidine 5'-monophosphate (dTMP), using the cosubstrate, 5,10- methylenetetrahydrofolate (CH2H4folate) as a 1-carbon donor and reductant and contributes to the de novo mitochondrial thymidylate biosynthesis pathway. {PubMed:11278511, PubMed:21876188}.

Background Thymidylate synthase (TS) is an intracellular enzyme critical for de novo synthesis of DNA. In cancer, expression of TS is often elevated and becomes further elevated as a result of treatment with the most commonly used chemotherapeutic, 5-fluorouracil (5-FU). Resistance or lack of response to 5-FU is attributed to the elevation of TS activity. Overexpression of TS has been demonstrated in many types of tumors including breast, colon, gastric and melanoma and is usually predictive of shortened survival time.

Storage This antibody is stable for at least one (1) year at -20°C. Avoid multiple freeze- thaw cycles.
Dilution Instructions Dilute in PBS or medium that is identical to that used in the assay system.
Application Instructions Immunohistochemistry: use at 5-10 ug/ml on frozen sections (reactivity on paraffin-embedded sections has not been determined)
Immunoblotting: use at 5-10 ug/ml
ELISA: use at 2-5 ug/ml (optimized for TS on solid phase at 5 ug/ml).
