Biological Function DNA glycosylase which prefers single-stranded DNA (ssDNA), or partially ssDNA structures such as bubble and fork structures, to double-stranded DNA (dsDNA) (PubMed:12433996, PubMed:19170771, PubMed:22569481, PubMed:23755964). Mediates interstrand cross-link repair in response to replication stress: acts by mediating DNA glycosylase activity, cleaving one of the two N-glycosyl bonds comprising the interstrand cross-link, which avoids the formation of a double-strand break but generates an abasic site that is bypassed by translesion synthesis polymerases (By similarity). In vitro, displays strong glycosylase activity towards the hydantoin lesions spiroiminodihydantoin (Sp) and guanidinohydantoin (Gh) in both ssDNA and dsDNA; also rUniProtKB:A0A1L8HU22, PubMed:12433996, PubMed:19170771, PubMed:22569481, PubMed:23755964}.
Research Areas DNA
Background Prokaryotes and lower eukaryotes possess redundant activities that remove the oxidative DNA base damages produced during normal oxidative metabolism and which have been associated with cancer and aging. Thus far, only one oxidized pyrimidine- specific DNA glycosylase has been identified in humans, hNthl. NEI3 is one of three new putative human DNA glycosylases.
Handling
Storage This antibody is stable for at least one (1) year at -20°C. Avoid multiple freeze- thaw cycles.
Dilution Instructions Dilute in PBS or medium which is identical to that used in the assay system.
Application Instructions Immunoblotting: use at 1:500 dilution. A band of approximately 68 kDa is detected.
Positive control: human testes tissue lysate.
Positive control: human testes tissue lysate.
