Specificity Information
Specificity This antibody reacts with the highly conserved heptapeptide repeat of the largest subunit of eukaryotic RNA polymerase II. It inhibits promoter- directed transcription, but it does not inhibit elongation in the nonspecific transcription assay.
Target Name DNA-directed RNA polymerase II subunit RPB1
Target ID RNA Polymerase II
Uniprot ID P24928
Alternative Names RNA polymerase II subunit B1, EC 2.7.7.6, DNA-directed RNA polymerase II subunit A, DNA-directed RNA polymerase III largest subunit, RNA-directed RNA polymerase II subunit RPB1, EC 2.7.7.48
Gene Name POLR2A
Sequence Location Nucleus, Cytoplasm, Chromosome
Biological Function DNA-dependent RNA polymerase catalyzes the transcription of DNA into RNA using the four ribonucleoside triphosphates as substrates. Largest and catalytic component of RNA polymerase II which synthesizes mRNA precursors and many functional non-coding RNAs. Forms the polymerase active center together with the sUniProtKB:P08775, PubMed:20231364, PubMed:23748380, PubMed:24207025, PubMed:26124092, PubMed:9852112}.; (Microbial infection) Acts as an RNA-dependent RNA polymerase when associated with small delta antigen of Hepatitis delta virus, acting both as a replicate and transcriptase for the viral RNA circular genome. {PubMed:18032511}.
Research Areas Protein Transcription
Handling
Storage These antibodies are stable for at least one (1) year at -20°C to -70°C. Store product in appropriate aliquots to avoid multiple freeze-thaw cycles.
Dilution Instructions Dilute in PBS or medium that is identical to that used in the assay system.
Application Instructions This antibody may be used to detect unproteolyzed RNA polymerase II or to inhibit specific transcription from class II promoters. This is a polyol-responsive antibody and can be used in gentle purifications of RNA polymerase II from from wheat germ, calf thymus, and yeast and is likely to purify RNA polymerase II from most eukaryotic organisms.
