Biological Function Component of NMDA receptor complexes that function as heterotetrameric, ligand-gated ion channels with high calcium permeability and voltage-dependent sensitivity to magnesium. Channel activation requires binding of the neurotransmitter glutamate to the epsilon subunit, glycine binding to the zeta subunit, plus membrane depolarization to eliminate channel inhibition by Mg(2+) (PubMed:1350383, PubMed:1834949, PubMed:1388270, PubMed:8428958, PubMed:18177891, PubMed:28384476, PubMed:15996549, PubMed:24876489, PubMed:27135925, PubMed:27618671, PubMed:28468946). Sensitivity to glutamate and channel kinetics depend on the subunit composition (PubMed:28384476). {PubMed:1350383, PubMed:1388270, PubMed:15996549, PubMed:18177891, PubMed:1834949, PubMed:24876489, PubMed:27135925, PubMed:27618671, PubMed:28384476, PubMed:28468946, PubMed:8428958}.
Research Areas Neuroscience
Background The NMDA receptor (NMDAR), a glutamate receptor, is the predominant molecular device for controlling synaptic plasticity and memory function. NMDAR is a heterotetramer of two NR1 and two NR2 subunits. Multiple receptor isoforms with distinct brain distributions and functional properties arise by selective splicing of the NR1 transcripts and differential expression of the NR2 subunits.
