Biological Function Calcium-binding protein that interacts with newly synthesized monoglucosylated glycoproteins in the endoplasmic reticulum. It may act in assisting protein assembly and/or in the retention within the ER of unassembled protein subunits. It seems to play a major role in the quality control apparatus of the ER by the retention of incorrectly folded proteins. Associated with partial T-cell antigen receptor complexes that escape the ER of immature thymocytes, it may function as a signaling complex regulating thymocyte maturation. Additionally it may play a role in receptor-mediated endocytosis at the synapse.

Background Calnexin is a ~90 kDa integral protein of the endoplasmic reticulum; it is also referred to as IP90, p88 and p90. It consists of a 50kDa N-terminal calcium- binding luminal domain, a single transmembrane helix, and a short acidic cytoplasmic tail. Its positively charged cytosolic residues ensure its retention in the endoplasmic reticulum. Calnexin together with calreticulin play a key role in glycoprotein folding by interacting with folding intermediates via their monoglycosylated glycans. Calnexin has also been shown to associate with the major histocompatability complex class I heavy chains, partial complexes of the T cell receptor, and B cell surface immunoglobulin.
Handling
Storage This product is stable for at least one (1) year at -20°C to -70°C. Reconstituted product should be stored in appropriate aliquots to avoid repeated freeze-thaw cycles.
Dilution Instructions Reconstitute lyophilized antibody using ultrapure to a final concentration no less than 100 ug/mL. Reconstitution with PBS or Tris buffer is acceptable if required. Protein carrier, biocide or cryopreservative can be added as needed. Aliquot and store at -20C for longterm storage. Dilute immediately prior to use.
Application Instructions Immunoblotting: use at 1-10ug/ml.
Immunohistochemistry: use at 1-10ug/ml.
