Research Areas Heat Shock& Stress Proteins
Background Grp170 is a major chaperone/stress protein resident in the ER. It is distantly related in sequence to both hsp110 and hsp70 families, but represents the Grp170 stress protein family. Studies have suggested that Grp170 is an important element of the protein processing machinery of the ER. Unlike Hsp110 and Hsp70, Grp170 has two distinct chaperoning domains; a classic beta-sheet peptide binding domain, with some COOH- terminal sequence, and a COOH-terminal alpha-helical domain. Like other members of the Hsp70 superfamily, Grp170 can elicit anti-tumor immune responses most likely by chaperoning antigenic peptide and binding to receptor(s) on antigen-presenting cells (APC).
Handling
Storage This antibody is stable for at least one (1) year at -20°C.
Dilution Instructions Dilute in PBS or medium that is identical to that used in the assay system.
Application Instructions ELISA: confirmed with antibody (1-10ug/ml) binding to Grp170 at 5ug/ml on the solid phase
Immunoblotting: with antibody at 1.0-2.0ug/ml. A band of 170kDa is detected.
These are recommended concentrations
End user should determine optimal concentrations for their applications.
Positive control: HEK293 cell lysate.
